SIĞIR VE KOYUNLARDAN ELDE EDİLEN FASCİOLA SPP İZOLATLARINDAKİ GENETİK ÇEŞİTLİLİĞİN MULTİPLEKS PCR VE SEKANS ANALİZİ İLE ARAŞTIRILMASI

Abstract

Fasciolosis, Fasciola hepatica ve F. gigantica'nın neden olduğu oldukça yaygın bir helmint enfeksiyonudur. Bu çalışmada fosfoenol piruvat karboksikinaz (pepck) genini amplifiye ederek hibrid Fasciola türlerini tanımlamak amacıyla bir multipleks PCR yapıldı. Ayrıca Fasciola haplotiplerini belirlemek için mitokondriyal nad1 amplifiye edilerek dizi ve filogenetik analizleri yapıldı. Bu amaçla, Elazığ ilindeki mezbahanelerden kesimi takiben 51 sığır ve 51 koyundan erişkin Fasciola izolatları toplandı, genomik DNA izole edildi ve ardından pepck'i çoğaltmak için tek aşamalı multipleks PCR yapıldı. 510 bp'lik bir bant oluşturan izolatlar F. gigantica, 241 bp'lik bir bant oluşturanlar F. hepatica ve her iki bandı oluşturanlar da hibrid (aspermik) parazitler olarak tanımlandı. Multipleks PCR neticesinde, dört izolat F. gigantica ve 84 izolat da F. hepatica olarak tanımladı. On dört de hibrid izolat (beş sığır ve dokuz koyun) tanımlandı. nad1 PCR ürünlerinin tek yönlü DNA dizi analizinde, üç izolatın F. gigantica ve 99 izolatın da F. hepatica olduğu teyid edildi. Ayrıca F. hepatica nad1 sekansları ile tespit edilen 87 haplotipten dördünün daha önce rapor edildiği, geri kalan 83'ünün de bu çalışmaya özgü olduğu belirlendi. Sonuç olarak, bu çalışma ile Türkiye'de ilk kez hibrid Fasciola parazitlerinin ve F. hepatica'nın daha önce tanımlanmamış 83 haplotipinin varlığı ortaya konuldu.Fasciolosis is a highly prevalent helminthic infection caused by Fasciola hepatica and F. gigantica. With the aim of identifying hybrid Fasciola flukes, multiplex PCR was performed to amplify phosphoenolpyruvate carboxykinase (pepck) gene. Furthermore, to determine Fasciola haplotypes, mitochondrial nad1 was amplified and aldo sequence and phylogenetic analyses were performed. Adult Fasciola isolates were collected from 51 cattle and 51 sheep after slaughtering from the abbatoires in Elazig province of Turkey, genomic DNA was isolated, and one-step multiplex PCR was subsequently performed for amplifying pepck. Isolates that generated a 510 bp band were identified as F. gigantica, those that generated a 241bp band were identified as F. hepatica, and those that generated both bands were identified as hybrid (aspermic) flukes. Further, multiplex PCR data identified four isolates as F. gigantica and 84 as F. hepatica. Fourteen hybrid isolates (five cattle and nine sheep) were identified. On unidirectional DNA sequence analysis of nad1 PCR products, three sequences were identified as F. gigantica and 99 as F. hepatica. Besides, four of 87 haplotypes de-tected for F. hepatica nad1 sequences were found to be previously reported, while the remaining 83 are unique to this study. To summarize, this study reports for the first time the existence of hybrid Fasciola flukes and 83 unique haplotypes of F. hepatica in Turkey.
