ISOPROTERENOL İLE DENEYSEL MİYOKARD ENFARKTÜSÜ OLUŞTURULAN SIÇANLARDA NEROLİDOL VE VİTAMİN D' NİN ETKİLERİNİN ARAŞTIRILMASI

Abstract

Kardiyovasküler hastalıklar küresel olarak önde gelen ölüm nedenlerinin başında gelmektedir. Akut miyokard enfarktüsü (MI) geçiren hastaların mortalite ve morbidite oranları genel popülasyona oranla 1.5 kat yüksektir. Nerolidol(NRD), çiçek kokulu çeşitli bitkilerin uçucu yağında bulunan doğal olarak oluşan bir seskiterpen alkoldür. Vitamin D(VİT D), metabolizmada başta kemik ve iskelet sistemi olmak üzere birçok önemli fonksiyona sahiptir. Yapılan araştırmalar NRD ve VİT D' nin hipertansiyon ve kardiyovasküler hastalıkların tedavisinde kullanılabileceği yönündedir. Çalışmamıza 42 adet 8-10 haftalık Wistar Albino erkek sıçanlar dahil edildi ve her grupta 7 sıçan olmak üzere 6 gruba ayrıldı. Gruplar; kontrol, MI, NRD, MI+NRD, VİT D, MI+VİT D şeklinde oluşturuldu. Akut MI oluşturmak için 200 mg/kg Isoproterenol kullanıldı. Tedavi için sıçanlara mısırözü yağında çözdürülen NRD(100 mg/kg/gün) ve VİT D (50 IU/gün) 14 gün boyunca uygulandı. NRDve VİT D' nin terapötik etkileri kardiyak belirteçler, anti-oksidan, biyokimyasal parametreler, histopatolojik ve immünohistokimyasal yöntemler kullanılarak incelendi. Çalışma sonucunda MI grubuna kıyasla MI+NRD ve MI+VİT D gruplarında total antioksidan seviyelerinde(TAS) artma ve total oksidan seviyelerinde(TOS) anlamlı bir azalma olduğu görüldü (p<0,05). Asprosin düzeylerinde, MI grubuna kıyasla MI+NRD ve MI+VİT D gruplarında anlamlı bir azalma tespit edildi (p<0,05). Yine MI grubu ile kıyaslandığında MI+NRD ve MI+VİT D gruplarında Meteorin Like Protein (METRNL) düzeylerinin ise anlamlı şekilde arttığı gözlendi (p<0,05). Histopatolojik incelemelerde MI+NRD ve MI+VİT D gruplarında MI grubuna oranla eritrosit ekstravazasyonu ve fibrozisin anlamlı düzeyde azaldığı ve nekrotik kas liflerinde anlamlı derecede iyileşme olduğu görüldü (sırası ile p<0,001, p=0,002). Nerolidol ve Vitamin D' nin MI tedavisinde etkili olduğu ve bu etkinin METRNL'e bağımlı ancak asprosinden bağımsız olduğu görüldü.Cardiovascular diseases are one of the leading causes of death globally. The mortality and morbidity rates of patients with acute myocardial infarction (MI) are 1.5 times higher than in the general population. Nerolidol (NRD) is a naturally occurring sesquiterpene alcohol found in the essential oil of various floral-scented plants. Vitamin D (VIT D) has many important functions in metabolism, especially in the bone and skeletal system. Studies have shown that NRD and VIT D can be used to treat cardiovascular diseases and hypertension. 42 numbers of 8-10 week old Wistar Albino male rats were included in our study, and they were divided into 6 groups with 7 rats in each group. Working groups include Control, MI, NRD, NRD+MI, VIT D, MI+VIT D. 200 mg/kg Isoproterenol was used to induce acute MI. Rats were treated with NRD dissolved in corn oil (100 mg/kg/day) and VIT D (50 IU/day) for 14 days. The therapeutic effects of NRD and VIT D were examined with cardiac markers, oxidation and biochemical parameters, and histopathological and immunohistochemical methods. As a result, MI+NRD and MI+VIT D groups, compared to the MI group, increased the total antioxidant status(TAS) and decreased the total oxidant status(TOS) level significantly (p<0,05). Asprosin levels were significantly lower in the MI+NRD and MI+VIT D groups compared to the MI group (p<0,05). Meteorin Like Protein (METRNL) levels significantly increased in the MI+NRD and MI+VIT D groups compared to the MI group (p< 0,05). In histopathological examinations, it was observed that erythrocyte extravasation, and fibrosis were significantly reduced, and necrotic muscle fibers showed a significantly improved pattern in the MI+NRD and MI+VIT D groups compared to the MI group. (p<0,001, p=0,002, respectively). Nerolidol and Vitamin D were found to be effective in the treatment of MI, and this effect was dependent on METRNL but independent of asprosin.
