SIÇAN AORT'UNDA KASILMA-GEVŞEME MEKANİZMASI ÜZERİNE NESFATİN-1'İN ETKİLERİ

Abstract

Nesfatin-1, son dönemlerde keşfedilen önemli endokrin ve metabolik işlevleri olduğu düşünülen muhtemel yeni bir metabolik hormondur. Bu bilgiler ışığında yapılan çalışmada 4-6 aylık 200-250 gram ağırlığındaki erkek Wistar cinsi sıçanlarda nesfatin-1'in sıçan aort kontraksiyonları üzerindeki olası etkisi araştırılmıştır. Çalışmada sıçan aortuna, 0.1, 1 ve 10 nM dozlarda kümülatif olmayan uygulamalarla nesfatin-1'in izometrik kontraksiyonlar üzerindeki doz bağımlı etkisi izole organ banyosu kullanılarak test edildi. Ayrıca bu uygulamanın sıçan aort dokusundaki değişimde immüno-histokimyasal boyama yöntemiyle histolojik olarak gösterildi. Çalışmanın sonucundanesfatin-1'in farklı dozlarda doz bağımlı olarak sıçan aort kontraksiyonlarını inhibe ettiği ve uygulamanın sıçan aortunda immüno-histokimyasal değişimlere neden olduğu gözlendi. Kasılma ve gevşeme gerilim yüzdeleri açısından 0.1 nM nesfatin uygulanan gurupta istatistiksel olarak herhangi bir farklılık oluşturmaz iken,1 nM ve 10 nM dozlarda ise istatiksel olarak anlamlı faklılıklar ortaya çıktı(p<0.05). 1 ve 10 nM doz uygulamasında aort kontraksiyon ve relaksasyonlarını kasılma ve gevşeme peryodunda istatistiksel olarak anlamlı derecede azalttığı tespit edildi (p<0.05 ). Çalışma sonucunda elde edilen bulgular nesfatin-1' in doz bağımlı olarak sıçan aort kontraksiyon ve gevşeme protokollerinin her ikisine de inhibisyon yaptığı gözlenmiştir ayrıca nesfatin uygulamasının doz bağımlı bu etkileri histolojik olarak doku düzeyinde de tespit edilmiştir.

Nesfatin-1 is likely to be a new metabolic hormone recently discovered which is thought to have the significant endocrine and metabolic functions. The study carried out in the light of this data attempts to find the effect of Nesfatin-1 on the aorta contractions of 4-6 month-old male Wistar rats weighing 200–250 g. In this study the dose-dependent effect of nesfatin given to the aorta of rats at doses of 0.1, 1 ve 10 nM non-cumulatively methods on isometric contractions has been tested by using isolated organ bath. In addition, the effect of this application on aortic tissue of rats has been histologically presented using histo-chemical staining methods. It has been revealed at the end of the study that nesfatin-1 at different doses inhibits the contractions of rat aorta in a dose dependent manner and this application results in immunohistochemical changes in rat aorta. In terms of the contraction and relaxation stress percentages it has not resulted in any statistical difference within the group at a dose of 0.1nM nesfatin while there is a statistically significant difference among the groups at doses of 1 nM and 10 nM(p<0.05). Nesfatin application at doses of 1 and 10 nM has been found to decrease the aortic contractions in a statistically significant way (p<0.05 ). The se results have demonstrated that nesfatin-1 inhibits both the aortic contractions and relaxation procedures of rats in a dose dependent manner. Moreover, these dose-dependent effects of nesfatin application have been histologically identified in tissue level, as well.
