SDS-PAGE YÖNTEMİNİN F. HEPATİCA VE F. GİGANTİCA E/S PROTEİNLERİNİN GÖSTERİLMESİNDEKİ ETKİNLİĞİ

Abstract

Amaç: Bu çalışmada F. Hepatica ve F.gigantica'nın tanımlanmış E/S proteinlerinin gösterilmesinde SDS-PAGE'in etkinliğinin araştırılması amaçlandı. Gereç ve Yöntem: % 10'luk ayırıcı jel (seperasyon jeli) ve % 5'lik toplayıcı jel kullandığımız SDS-PAGE ile E/S proteinlerini ayrıştırdık. Bulgular: Çalışmamızda F. Hepatica E/S proteinlerinden bilinen 30'a yakın proteinden yaklaşık 20'sini saptarken F. gigantica E/S proteinlerinin bilinen 6 proteininden ancak 2'sini saptadık. Sonuç: F. Hepatica E/S proteinlerinin gösterilmesinde % 10'luk jel kullanılan SDS-PAGE yöntemi uygun bulunurken F. gigantica için aynı başarı sağlanamamış ve yöntemde modifikasyonlar gerekebileceği kanısına varılmıştır.

Aim: The aim of this study was to investigate the efficiency of SDS-PAGE in visualization of described E/S proteins. Materials and Methods: SDS-PAGE seperation was performed by using 10% seperation gel and 5% stacking gel. Results: About 20 out of the 30 known F. Hepatica E/S proteins and 2 out of 6 F. gigantica E/S proteins were determined. Conclusion: Use of 10% gel in SDS-PAGE method was efficient in determination of F. Hepatica E/S proteins but the success rate was not the same for determination of F. gigantica.
