DONDURULMUŞ BOĞA SPERMATOZOONLARININ PROGRESSİF MOTİLİTESİ, KİNEMATİK PARAMETRELER VE MİTOKONDRİYAL MEMBRAN POTANSİYELİ ÜZERİNE ÇÖZDÜRME SONRASI FARKLI SICAKLIK VE BEKLETME SÜRELERİNİN ETKİLERİ

Abstract

Bu çalışmanın amacı, dondurulmuş boğa spermasının çözdürme sonrası yüksek mitokondriyal membran potansiyeli, motilite ve kinematik hız parametre değerleri üzerine farklı inkübasyon süreleri ve sıcaklık derecelerinin etkisini araştırmaktı. Çalışmada dört farklı boğanın 0,25 ml'lik payetlerdeki dondurulmuş sperma örnekleri kullanıldı. Payetler, 38°C'deki su banyosunda 25 saniye boyunca çözdürüldü. Çözdürüldükten sonra, bir CASA sistemi kullanılarak sperm motilitesi ve kinematik parametreler belirlendi. Sperm mitokondriyal membran potansiyeli ise JC-1 floresan ayracı kullanılarak flow sitometri ile incelendi. Çözdürüldükten sonra farklı sıcaklık derecelerine (35°C, 22°C ve 5°C) ve inkübasyon sürelerine (hemen, 10 ve 20 dakika) maruz bırakılan sperma örneklerinin motilite, sperm kinematik parametreleri ve yüksek mitokondriyal membran potansiyeli araştırıldı. Boğalar arasında çözdürme sonrası total ve progresif motilitesi açısından bireysel olarak önemli farklılıkların (p<0,05) olduğu gözlendi. Farklı inkübasyon sürelerinde 35°C ve 22°C arasında tüm kriterler açısından anlamlı bir fark gözlenmedi (p?0,05). Öte yandan, çözdürme sonrası 5°C'deki inkübasyonun 10. ve 20. dakikalarında sperm toplam ve progresif motilitesi ile VCL, VAP ve VSL gibi sperm kinematik hız parametreleri önemli ölçüde (p<0,05) azaldı. Benzer şekilde, yüksek mitokondriyal membran potansiyeline sahip spermatozoon oranı da 5°C'de, diğer sıcaklık dereceleriyle karşılaştırıldığında aynı zaman periyotları içinde önemli derecede azaldı. Ayrıca, progresif motilite, VCL, VAP, VSL ve yüksek mitokondriyal membran potansiyeli arasında yüksek bir pozitif korelasyon tespit edildi (p<0,01). Sonuç olarak, çözdürme sonrası spermatozoonların 5°C gibi düşük sıcaklık ortamında 20 dakika inkübasyon süresine maruz bırakılması; sperm motilitesi, sperm kinematik hız parametreleri ve yüksek mitokondriyal membran potansiyelini olumsuz yönde etkilenmiştir.The aim of the present study was to investigate the effect of different incubation times and thermal conditions on sperm high mitochondrial membrane potential, sperm motility and kinetic parameters after thawing in frozen bull semen. In the study, frozen semen samples, in straws of 0,25 ml, from four different bulls were used. Straws were thawed for 25 seconds in water baths at 38°C. After thawing, the sperm motility and kinetic parameters determined using a CASA system and the sperm mitochondrial membrane potential examined using JC-1 by flow cytometry. Total and progressive motility, sperm kinetic parameters and high mitochondrial membrane potential were investigated in different thermal conditions (35°C, 22°C and 5°C) and incubation times (immediately, 10 and 20 minutes) after thawing. Significant individual differences (p<0.05) were observed between bulls in terms of total and progressive motility after thawing. No significant difference (p?0.05) was observed with respect to all criteria between 35°C and 22°C at different incubation times. On the other hand, the sperm total and progressive motility, the sperm kinematics as VCL, VAP and VSL significantly reduced (p<0.05) at 10 and 20 minutes after thawing in 5°C. Similarly, the proportion of spermatozoa with high mitochondrial membrane potential also decreased (p<0.05) significantly at 5°C during the same time periods compared to other temperatures. There was a high correlation among progressive motility, VCL, VAP, VSL and high mitochondrial membrane potential (p<0.01). In conclusion, sperm motility, sperm kinetic parameters and high mitochondrial membrane potential negatively affected during the incubation period of 20 minutes after thawing in a low-temperature environment such as 5°C.
