RATLARDA GÜRÜLTÜNÜN OLUŞTURDUĞU OKSİDATİF STRESE KARŞI E VİTAMİNİN KORUYUCU ETKİSİ

Bu çalışmada, gürültünün oluşturduğu oksidatif strese karşı, önemli bir antioksidan olan E vitamininin koruyucu etkisinin araştırılması amaçlanmıştır. Bu amaçla, 21 Wistar-Albino ırkı erkek sıçanlardan, her grupta 7 adet olacak şekilde üç grup oluşturuldu. Bu gruplar; 1. Grup: Kontrol grubu olarak sadece plasebo (mısır yağı) uygulandı. 2. Grup (gürültü): Sıçanlar, 30 gün boyunca günde 10 saat, 8.000-9.000 hertz ve 100 desibel şiddetindeki sese maruz bırakıldı. Plasebo olarak mısır yağı uygulandı. 3. Grup (Gürültü+E vitamini): Sıçanlara, gürültüye ilaveten E vitamini (200 mg/kg/gün) mısır yağında çözdürülerek, ağızdan gavaj yoluyla verildi. E vitamini gürültü maruziyetinden bir hafta önce başlandı. Daha sonra, gürültü maruziyetinden 2 saat önce 30 gün süreyle verildi. Çalışma sonunda, sıçanlar yaklaşık 12 saat açlığı müteakiben anestezi edilerek, kan ve doku örnekleri alındı. Elde edilen eritrosit ve doku örneklerinden malondialdehit (MDA), indirgenmiş glutatyon (GSH) düzeyleri, glutatyon peroksidaz (GSH-Px) ve katalaz aktiviteleri belirlendi. Elde edilen sonuçlara göre, eritrosit ve karaciğer MDA düzeylerinde gürültünün etkisine bağlı olarak 2. grupta önemli düzeyde artış (p<0,05) olduğu, buna karşın E vitamini verilen 3. grupta bu artışın önemli düzeyde azaldığı (p<0,05) tespit edildi. Kontrol ile E vitamini verilen grup arasında MDA düzeyinde bir farkın olmaması, E vitamininin koruyucu etkisini ortaya koymaktadır. Gürültüye maruz kalan grupta eritrosit GSH ve GSH-Px aktivitesinde bir fark olmamasına rağmen, karaciğerde önemli bir azalma (p<0,05) saptandı. Ayrıca E vitamini verilen grupta eritrosit ve karaciğer GSH ile karaciğer GSH-Px'te önemli bir artış (p<0,05) belirlendi. Gürültü stresinin uygulandığı grupta eritrosit ve karaciğer katalaz aktivitesinde önemli bir azalma (p<0.05) olmasına rağmen, E vitamini uygulanan grupta önemli bir artış (p<0.05) tespit edildi. Böbrek dokusunda, bütün parametrelerde gruplar arasında anlamlı bir fark belirlenmedi. Çalışma bulgularına göre, eritrosit ve karaciğerde gürültü maruziyetinden dolayı oksidatif stres göstergesi olan MDA'nın arttığı ve antioksidan parametrelerin azaldığı saptanmıştır. Buna karşın, E vitamini MDA düzeylerini azaltırken, eritrosit ve karaciğer GSH düzeyini ve GSH-Px ve katalaz aktivitelerini arttırarak koruyucu bir etki göstermiştir. Sonuç olarak; gürültü dokularda oksidatif stresi arttırırken, E vitamininin gürültü stresine karşı önemli bir koruyucu etki gösterdiği tespit edilmiştir.
In this study, it was aimed to investigate the protective effect of vitamin E, which is an important antioxidant, against the oxidative stress that noise creates. For, this purpose, 21 male Wistar-Albino rats divided to 3 group, 7 in each group. These groups are; Group 1: As a control group, only placebo (corn oil) was implemented. Group 2 (noise): Rats were exposed to 8,000-9,000 hertz and 100 decibels intensity of voice for 10 hours a day for 30 days. Corn oil was administered as a placebo. Group 3 (Noise + Vitamin E): Rats were given Vitamin E (200 mg/kg/day) in corn oil by oral gavage in addition to noise. Vitamin E was implemented a week before noise exposure. Then, it was given 2 hours before noise exposure for 30 days. At the end of the study, rats were anesthetized at the end of fasting for about 12 hours and blood and tissue specimens were taken. Malondialdehyde (MDA), reduced glutathione (GSH) levels, glutathione peroxidase (GSH-Px) and catalase activities were determined from the obtained erythrocytes and tissue specimens. According to the results, erythrocyte and liver MDA levels were significantly increased in group 2 (p<0.05) due to the effect of noise, whereas in the third group given vitamin E, this increase was significantly decreased (p<0.05). The absence of a difference in the level of MDA between the control and the vitamin E-treated group indicates the protective effect of vitamin E. A significant decrease (p<0.05) in the liver was detected, although there was no difference in the activity of erythrocyte GSH and GSH-Px in the group exposed to noise. A significant increase in erythrocyte and hepatic GSH and hepatic GSH-Px (p <0.05) was also observed in the group given E vitamins. A significant increase (p<0.05) was noted in the group receiving E vitamins, although there was a significant decrease (p<0.05) in the activity of erythrocyte and liver catalase in the group in which noise stress was applied. There was no significant difference between groups in all parameters in kidney tissue. According to the findings of the study, it was determined that due to noise exposure in erythrocyte and liver MDA which is an oxidative stress indicator is increased and antioxidant parameters are decreased. While vitamin E reduced MDA levels, on the other hand, it showed a protective effect by increasing erythrocyte and liver GSH levels and GSH-Px and catalase activities.
