| dc.description.abstract | Metabolik sendrom; abdominal obezite, insülin direnci, dislipidemi ve hipertansiyonun birlikte seyrettiği, kardiyovasküler hastalık ve tip 2 diyabet riskini belirgin biçimde artıran kompleks bir klinik tablodur. Güncel çalışmalar, fruktozdan zengin diyetlerin hepatik steatoz, oksidatif stres ve kronik düşük dereceli inflamasyon yoluyla metabolik sendrom gelişimini hızlandırdığını göstermektedir. Bitkisel kaynaklı çoklu doymamış yağ asitleri ve fenolik bileşenler içeren yağların bu süreçler üzerinde koruyucu veya modülatör etkiler gösterebileceği bildirilmektedir. Keten tohumu yağı, alfa-linolenik asit (?-3), lignanlar ve fenolik bileşikler bakımından zengin olan keten tohumu yağı, güçlü antioksidan ve antiinflamatuvar özellikleri nedeniyle dikkat çekmektedir. Bu tez çalışmasının amacı, yüksek fruktoz diyeti ile metabolik sendrom benzeri tablo oluşturulan Sprague-Dawley ırkı dişi sıçanlarda keten tohumu yağının karaciğer ve beyin üzerindeki metabolik, oksidatif ve inflamatuvar etkilerini; özellikle mGluR2, mGluR5, iNOS ve COX-2 protein düzeyleri bağlamında değerlendirmektir. Sprague-Dawley ırkı albino dişi sıçanlar 12 hafta boyunca kontrol, keten tohumu yağı (KTY), yüksek fruktoz diyeti (YFD) ve KTY+YFD gruplarına ayrılmıştır. Deneme sonunda serum glukoz, trigliserit, üre, kreatinin, ALT ve AST düzeyleri ölçülmüş; antioksidan yanıt serum katalaz (CAT) ve glutatyon peroksidaz (GPx) aktiviteleri üzerinden değerlendirilmiştir. Karaciğer ve beyin dokularında mGluR2, mGluR5, iNOS ve COX-2 protein düzeyleri Western blot ile analiz edilmiştir. Veriler tek yönlü ANOVA ve Tukey testi ile değerlendirilmiş, p<0.05 anlamlı kabul edilmiştir. Canlı ağırlık değişimi anlamlı olup (p<0.001), karaciğer ve visseral yağ ağırlıkları (p<0.001) ile serum glukoz (p<0.001) ve trigliserit (p<0.01) düzeyleri kontrol grubuna kıyasla artarken, KTY takviyesi bu artışları düşürmüştür (p<0.05). ALT ve AST aktiviteleri YFD grubunda kontrole kıyasla artarken (p<0.001), KTY+YFD grubunda ise YFD'ye kıyasla azalmıştır (p<0.05). CAT ve GPx aktiviteleri YFD'ye kıyasla KTY takviyesiyle artmıştır (p<0.05). Western blot analizleri, YFD grubunda karaciğer ve beyinde mGluR2, mGluR5, iNOS ve COX-2 ekspresyonlarının arttığını (p<0.05), KTY+YFD grubunda ise bu artışın anlamlı biçimde baskılandığını göstermiştir (p<0.05). Sonuç olarak, keten tohumu yağı yüksek fruktoz diyetine bağlı olarak gelişebilen kilo alımı modelinde metabolik bozulmaları kısmen iyileştiren, oksidatif stres ve inflamasyonu azaltan ve karaciğer ile beyinde glutamaterjik/inflamatuvar yanıtı modüle edebilen tamamlayıcı bir besinsel ajan olarak değerlendirilebileceği düşünülmektedir. | |
| dc.description.abstract | Metabolic syndrome is a complex clinical condition characterized by the coexistence of abdominal obesity, insulin resistance, dyslipidemia, and hypertension, significantly increasing the risk of cardiovascular disease and type 2 diabetes. Current studies indicate that fructose-rich diets accelerate the development of metabolic syndrome through hepatic steatosis, oxidative stress, and chronic low-grade inflammation. Oils containing plant-derived polyunsaturated fatty acids and phenolic compounds have been reported to exert protective or modulatory effects on these processes. Flaxseed oil, rich in alpha-linolenic acid (?-3), lignans, and phenolic compounds, has attracted considerable attention due to its strong antioxidant and anti-inflammatory properties. The aim of this thesis was to evaluate the metabolic, oxidative, and inflammatory effects of flaxseed oil on the liver and brain in female Sprague-Dawley rats in which a metabolic syndrome-like condition was induced by a high-fructose diet, with particular emphasis on mGluR2, mGluR5, iNOS, and COX-2 protein levels. Female rats were divided into control, flaxseed oil (FSO), high-fructose diet (HFD), and FSO+HFD groups for 12 weeks. At the end of the experiment, serum glucose, triglyceride, urea, creatinine, ALT, and AST levels were measured, and antioxidant response was assessed through serum catalase (CAT) and glutathione peroxidase (GPx) activities. Protein levels of mGluR2, mGluR5, iNOS, and COX-2 in liver and brain tissues were analyzed using Western blot, and data were evaluated by one-way ANOVA followed by Tukey's test, with p<0.05 considered statistically significant. In the HFD group, body weight (p<0.001), liver and visceral fat weights (p<0.001), and serum glucose (p<0.001) and triglyceride levels (p<0.01) were significantly increased compared to the control group. Flaxseed oil supplementation significantly reduced these increases (p<0.05). ALT and AST activities were markedly elevated in the HFD group (p<0.001) but decreased in the FSO+HFD group compared to HFD (p<0.05). CAT and GPx activities increased following flaxseed oil supplementation (p<0.05). Western blot analyses revealed that mGluR2, mGluR5, iNOS, and COX-2 expression levels were elevated in the liver and brain of the HFD group (p<0.05), whereas this increase was significantly suppressed in the FSO+HFD group (p<0.05). In conclusion, flaxseed oil may be considered a complementary nutritional agent capable of partially improving metabolic disturbances induced by a high-fructose diet, reducing oxidative stress and inflammation, and modulating glutamatergic and inflammatory responses in the liver and brain. | |
