EGZERSİZ UYGULANAN RATLARDA KAS DOKUSU FOXOL/PGC-1? SİNYAL YOLAĞI ÜZERİNE BETA-KRİPTOKSANTİN'İN ETKİSİ

dc.description.abstractEgzersiz sırasında üretilen aşırı ROS, oksidatif stresin oluşmasına ve bu da fazla egzersiz yapanlarda kas yorgunluğu ve kas hasarının gelişmesine sebep olmaktadır. Peroksizom proliferatör ile aktive edilen reseptör ? ko-aktivatör 1-? (PGC-1?) proteini, lipit peroksidasyonu ve mitokondriyal biyogenez gibi bazı hücresel süreçlerde rol alan bir transkripsiyon faktörüdür. Son zamanlarda Forkhead-Box O sınıfı proteinlerin (FoxO'lar) mitokondriyal oksidatif stresi düzenlemek için PGC-1? ile birlikte işlev gördüğü gösterilmiştir. FoxO1/PGC-1? aktivasyonunun, mitokondriyal disfonksiyonu önleyerek hem in vivo hem de in vitro ROS birikimini azalttığı bildirilmiştir. Güçlü antioksidan özellikte olduğu bilinen ?-Kriptoksantin (BCX), bazı narenciye meyvelerde bulunan bir pro-vitamin A karotenoididir. Çalışmamızda, ?-Kriptoksantin takviyesinin, egzersiz performansı üzerine etkilerinin aydınlatılması ve FOXO1/PGC-1? sinyal yolağı üzerine potansiyel etkisinin belirlenmesi amacıyla 28 adet 8 haftalık erkek Wistar Albino cinsi ratlar kullanılmıştır. Uygulanan egzersiz motorlu kemirgen koşu bandında, haftada beş gün 25m/45 dk şeklinde gerçekleştirilmiştir. BCX gruplarına günlük 100 ?g/kg oral gavaj yoluyla uygulama yapılmıştır. Laboratuvar analizleri sonucunda, BCX uygulanan gruplarda oksidatif hasarın göstergesi olan MDA seviyelerinde kontrol grubuna kıyasla düşüş gözlemlenmiştir (P<0.05). Üstelik bu düşüşün BCX uygulaması ile birlikte egzersiz uygulanan gruplarda arttığı görülmüştür. Süperoksit dismutaz (SOD), katalaz (CAT) ve glutatyon peroksidaz (GSH-Px) değerlerinde ise kontrole kıyasla diğer uygulama gruplarında artış gözlemlenmiştir. Yapılan ölçümlerde, PGC-1? protein seviyesinde kontrole oranla özellikle egzersiz ve BCX+egzersiz uygulama gruplarında ciddi bir artış gözlemlenmiştir (P?0.05). FOXO1 protein düzeyinin ise, hem BCX uygulama grubunda hem de BCX+egzersiz grubunda azalma gözlemlenmiştir (P<0.05). Çalışmamız sonucunda elde ettiğimiz veriler; BCX'in egzersiz performansı üzerine olumlu etkilerinin olduğunu ve egzersiz sonucu oluşabilecek olumsuz etkileri önlemek amacıyla kullanılabileceğini düşündürmektedir.
dc.description.abstractExcessive ROS produced during exercise causes oxidative stress to occur and this causes muscle fatigue and muscle damage in those who exercise excessive exercise. The receptor activated by peroxisoma proliferator is a transcription factor that takes part in some cellular processes such as co-active 1-? (PGC-1?) protein, lipid peroxidation and mitochondrial biogenesis. Recently, Forkhead-Box O class proteins (FoxOs) have been shown to function together with PGC-1? to regulate mitochondrial oxidative stress. It has been reported that FoxO1/PGC-1? activation prevents mitochondrial dysfunction and reduces ROS accumulation both in vivo and in vitro. ?-Cryptoxanthin (BCX), known to be strong antioxidant properties, is a pro-vitamin A carotenoid found in some citrus fruits. In our study, 28 8-week male Wistar Albino rats were used to illuminate the effects of ?-criptoxanthine supplement on exercise performance and to determine the potential effect on the FOXO1/PGC-1? signal path. The exercise was carried out on a motorized rodent treadmill, five days a week for 25m / 45 minutes. 100 ?g/kg oral gavage was applied to BCX groups daily. As a result of laboratory analysis, a decrease was observed in MDA levels, which is an indicator of oxidative damage, in BCX applied groups compared to the control group (P<0.05). Moreover, it was observed that this decrease increased in the groups that exercised together with the BCX application. As a result of laboratory analyzes, BCX decreased at MDA levels, which is indicative of oxidative damage in groups undergoing BCX, compared to the control group (p <0.05). Moreover, it has been seen that this decrease has increased in exercise groups with the BCX application. Superoxide dysmutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-PX) values were increased in other administration groups compared to control. In the measurements, a significant increase in exercise and BCX+exercise application groups were observed in the PGC-1? protein level (p?0.05). The FOXO1 protein level decreased both in the BCX application group and the BCX+exercise group (p <0.05). The data we obtained as a result of our study; It suggests that BCX has positive effects on exercise performance and can be used to prevent negative effects that may occur as a result of exercise.
