EFFECTS OF METOCLOPRAMIDE AND HYOSCINE–N-BUTLY BROMIDE ON DUODENUM AND COLON CONTRACTION-RELAXATION MECHANISM

| dc.description.abstract | Metoklopramid, kusma ile gastroenterit tedavisinde sıklıkla kullanılmaktadır. Hyoscine-N-butyl bromide (buscopan), asetilkolinin muskarinik reseptörler üzerindeki etkilerini bloke eden antikolinerjik bir ajandır. Bu çalışmanın amacı metoklopramidin asetilkolin ile indüklenen duodenum ve kolon hareketliliği üzerindeki etkisini belirlemekti. Diğer bir amaç ise metoklopramidin sadece ve hyosin-n-butylbromide ile duodenum ve kolon motilitesi üzerindeki etkisini araştırmaktır. Bu incelemede duodenum ve kolondan 1 cm uzakta, çıkarılmış ve 2 cm uzunluğunda şeritler hazırlanmış ve her iki tarafı cerrahi ipek ile sabitlenerek izole edilmiş organ banyosu dokusunun her iki ucunda (1.0 g) gerilim ile cihaza askıya alınmıştır. ve kaydedilen izometrik kasılmalar. Oniki parmak bağırsağına asetilkolin (10-4 M) eklenmiştir. 10 dakika sonra (35 uM) metoklopramid, (15 uM) hiyosin-N-butil bromid eklendi ve (50 uM) metoklopramid ve hiyosin-N-butil bromid üretildi ve inkübe edildi. Bu sonuçta (35 uM) metoklopramid, (15 uM) hiyosin-N-butil bromid ve (50 uM) metoklopramid ve hiyosin-N-butil için, duodenum ve kolonda frekans, tepeden tepeye ve genlik gösterildi. Daha sonra, sonuçların istatistiksel analizi, bir çift örnek T testi şeklinde gerçekleştirilir. Sonuç olarak, duodenumda, sadece metoklopramidin ve hiyosin-n-butil bromür ile etkinliği azalmış hareketlilik gösterir, ayrıca asetilkolini bloke eder. | dc.description.abstract | EFFECTS OF METOCLOPRAMIDE AND HYOSCINE–N-BUTLY BROMIDE ON DUODENUM AND COLON CONTRACTION-RELAXATION MECHANISM Metoclopramide is frequently used in the treatment of gastroenteritis with emesis. Hyoscine-N-butyl bromide (buscopan) is an anticholinergic agent that blocks the effects of acetylcholine on muscarinic receptors. The aim of this studied was to determine the effect of the metoclopramide upon acetylcholine-induced duodenum and colon motility. Another goal investigate the effect of metoclopramide only and with hyosin-n-butylbromide on duodenum and colon motility. In this examination 1 cm from the duodenum and colon, were removed and 2 cm long strips were prepared and fixed with both sides of surgical silk is fixed at both ends of isolated organ bath tissue with (1.0 g ) of tension to the device is suspended and recorded isometric contractions. In duodenum, were added acetylcholine (10-4 M). After 10 minutes, were added (35 µM) metoclopramide, (15 µM) hyoscine-N-butyl bromide, and (50 µM) metoclopramide and hyoscine-N-butyl bromide was produced and incubated. While, those are (35 µM) metoclopramide, (15 µM) hyoscine-N-butyl bromide, and (50 µM) metoclopramide and hyoscine-N-butyl in this result were showed frequency, peak to peak and amplitude in the duodenum and colon. Thereafter, statistical analysis of the results is carried out in the form of a paired sample T-test. In conclusion, in duodenum, the efficacy of metoclopramide only and with hyosin-n-butyl bromide the result shows reduced motility, also blocked acetylcholine. In colon, the effect of metoclopramide only and with hyosin-n-butyl bromide the result determined the increased motility also induced acetylcholine. | |
