BROYLERLER'DEN CLOSTRİDİUM PERFRİNGENS İZOLASYONU VE MOLEKÜLER KARAKTERİZASYONU

Abstract

Bu çalışmada, Elazığ ve Malatya illerindeki 100 farklı kümesten 3 farklı kesimhaneye getirilen broyler'lerde Clostridium perfringens'in varlığı konvansiyonel kültür ve PZR yöntemleri ile araştırılarak, söz konusu etkenin bölgedeki yaygınlığı ve dağılımı ile ilgili verilerin toplanması amaçlandı. Bu amaçla hayvanların büyüme dönemlerinde Malatya'da bulunan bir işletmeye ait 30 kümes ziyaret edildi. Malatya ve Elazığ'daki kanatlı kesimhanelerinden alınan 500 bağırsak ve 500 karaciğer örnekleri olmak üzere toplam 1000 numune incelendi. Alfa- toksin (?), beta-toksin (?), epsilon-toksin (?), iota-toksin (?), enterotoksin (Cpe), beta2 (?2) toksin genlerine spesifik primerler kullanılarak multiplex-PZR yöntemi ile izolatların toksin tiplendirmesi yapıldı. Ayrıca izolatlar, hasta ve sağlıklı kanatlı hayvanlardaki rolü henüz ortaya konulamamış olan NetB toksin geni yönünden PZR yöntemi ile incelendi.Elazığ ve Malatya'daki farklı kümeslerden izole edilmiş 61 izolata Pulsed Field Gel Electrophoresis (PFGE) yöntemi uygulanarak genotiplendirme yapıldı. Gel Compare II yazılım sistemi (version3.0; Applied Maths, Sint-Martens-Latem, Belgium) kullanılarak bant profilleri analiz edildi. PFGE profillerinin, dendogramı oluşturularak kümeleşme analizi yapıldı.Bu çalışmada Malatya ve Elazığ illerindeki broyler piliçlerde karaciğer ve ince bağırsaklarında toplam C. perfringens yaygınlığı %66 olarak tespit edildi. Toplam 660 C. perfringens izolatın 657'si ? toksin geni yönünden pozitif bulunurken, diğer toksin genlerinin hiçbirine rastlanmadı. Kalan üç örnek ise tiplendirilemedi. Çalışmada incelenen hiçbir izolatta NetB toksin geni saptanamadı. Kümesler arasında 30 farklı PFGE genotipi tespit edildi ve heterojen bir dağılım gözlendi. Bu çalışma ile Türkiye'de ilk kez kanatlı hayvanlarda hem NetB toksin geni araştırılmış, hem de C. perfringens izolatlarının PFGE profilleri saptanmıştır.

The aim of the current study was to investigate the prevalance of Clostridium perfringens in broilers which was brought for slaughter to 3 different slaughterhouses from 100 different broiler flocks in Elazig and Malatya provinces by conventional culture and PCR techniques. For this aim, 30 coops which were located in Malatya were visited during the growing period of broilers. A total of 500 intestines and 500 livers have been collected. Toxin genotyping were performed by using multiplex-PCR to the Alfa-toxin (?), beta-toxin (?), epsilon-toxin (?), iota- toxin (?), enterotoxin (Cpe), ?2 toxin genes. Besides all isolates were analysed for the NetB toxin gene using PCR. Also, a total of 61 isolates collected from different coops in Elazig and Malatya provinces were genotyped by Pulsed Field Gel Electrophoresis (PFGE) technique. Band profiles were analysed using the Gel Compare II software (version3.0; Applied Maths, Sint-Martens-Latem, Belgium). The dendogram of PFGE profiles was established and clustering analysis was performed. The prevalance of C. perfringens was detected as 66% in broilers located in Elazig and Malatya provinces. 657 out of 660 C. perfringens isolates was detected as positive for Alfa-toxin (?) while none of the other toxin genes could identified. It has not been genotyped the rest 3 samples. NetB toxin gene couldn't detected in none of the isolates examined. Thirty different PFGE genotypes were detected and a heterogeneous distribution was seen. Both NetB toxin gene and the PFGE profiles of C. perfringens isolates were investigated in broilers for the first time in Turkey in the current study.
