FARKLI SEVİYEDE ARSENİK İÇEREN YURTBAŞI ŞİFALI SULARINI İÇEN RATLARDA BİYOKİMYASAL VE HEMATOLOJİK PARAMETRELERDEKİ DEĞİŞİMLERİNİN İNCELENMESİ

Abstract

Arsenik, doğal olarak oluşabilen bir element olup, toprak ve suda hem organik hem de inorganik formlarda bulunabilir. Bu çalışma, Yurtbaşı (Elazığ)'nda bulunan ve insanlar tarafından çeşitli hastalıklara şifa dağıttığı gerekçesiyle içilen suyun, kısa dönemde canlılar üzerine etkilerini incelemek için planlanmıştır. Bu çalışmada iki ayrı derin kuyuda 1.000 ppb ve 15.000 ppb arsenik konsantrasyonları saptanmıştır. 6-8 haftalık olan ratlar 7'şer adetten üç gruba ayrılmıştır. Birinci grup normal içme suyu, ikinci grup 1.000 ppb arsenikli su, üçüncü grup ise 15.000 ppb arsenikli su ile 6 hafta boyunca beslenmiştir. 6. haftanın sonunda hayvan stresinden kaçınmak için anestezi olmadan servikal dekapitasyonla hayvanlar öldürülmüştür. İkinci ve üçüncü grup ratlarda ürik asit düzeylerinin, birinci grup ratlara göre istatistiksel olarak anlamlı şekilde artmış olduğu görüldü (p=0,04).İkinci ve üçüncü grup ratların plazma glukoz düzeylerinde anlamlı bir azalış gözlendi (p=0,005). İkinci ve üçüncü grup ratlarda, MCHC (p<0,001) ve EO (%,#) değerlerinin de (p=0,034, p=0,038 sırası ile ) arttığı görüldü. Arseniğin hedef organı olan karaciğer değerlerinde ciddi bir değişiklik görülmemiş olup bu da arsenik toksititesinin daha uzun bir süre sonra kronikleşebileceğini veya arseniğin dokulardan çabuk eliminasyona uğrayıp atıldığını göstermektedir. İkinci ve üçüncü grup ratlarda, MCHC ve eozinofil sayısındaki artış, demir eksikliği anemisine ve As toksititesine, bu da kan hücrelerinin erken kaybına işaret edebilir. Yine, yüksek dozlarda As alan ratlarda hiperürisemi görülmesi, protein katobolizmasına ait enzimlerde bir inhibisyon oluştuğunu göstermektedir. Dolayısıyla, yüksek düzeylerde As' li suların uzun süre tüketilmesi ya da bunların diğer canlılarda temel içme suyu olarak kullanılması sonucu, hipertansiyon, metabolik sendrom, koroner kalp hastalığı, oksidatif stres gibi patolojik bozukluklara yol açmaktadır. Bu tür içme suyu kullananlara arseniğin toksik düzeylerinden korunmak için, antioksidan moleküllerin daha fazla tüketilmesi önerilmektedir.Arsenic (As) is a naturally occurring element that can be found in both organic and inorganic forms in soil and water. This study was planned to investigate the effects of drinking water (with arsenic) in Yurtbaşı (Elazığ) on the grounds that it has been curing various diseases for a long time. In this study, the waters with arsenic concentrations of 1000 ppb and 15,000 ppb were collected from two deep wells. The rats, 6-8 weeks of age, were divided into three groups of seven. The first group was fed with tap water, the second group was fed with 1000 ppb arsenic water and the third group was fed with 15.000ppb arsenic water for 6 weeks. At the end of the 6th week, animals were sacrificed by cervical decapitation without anesthesia to avoid animal stress.Plasma Glucose levels were decreased and Uric acid levels were significantly increased in the second and third group rats compared to the first group rats (p=0.04) .MCHC (p<0,001) and EO (%, #) values (p=0.034, p=0,038 respectively). were increased in the second and third groups. The target organ of arsenic is the liver. There was no significant change in liver values.This suggests that arsenic toxicity may become chronic after a longer period of time, or that arsenic is rapidly eliminated from the tissues. MCHC and eosinophil count, iron deficiency anemia and As toxicity in second and third group rats. Therefore, this may indicate premature loss of blood cells. Again, the presence of hyperuricemia in rats receiving high doses of As shows an inhibition of protein catabolism enzymes. Thus, high levels of As waters are consumed for a long time or used as basic drinking water in other organisms, leading to pathological disorders such as hypertension, metabolic syndrome, coronary heart disease, oxidative stress. In order to protect the toxic levels of arsenic, users of this type of drinking water are advised to consume more antioxidant molecules.
