OKSİDE VE REDÜKTE GLUTATYON MİKTARLARI İLE GLUTATYON REDÜKTAZ VE PEROKSİDAZ ARASINDAKİ İLİŞKİNİN ARAŞTIRILMASI

Abstract

Bu çalışmada tip 2 diyabet teşhisi almış 60 kişi ile kontrol grubu olarak sağlık problemi olmayan 21 kişinin eritrosit, glutatyon peroksidaz (GSH-Px) ve glutatyon redüktaz (GSH-Rd) enzim aktivitesi UV-spektrofotometresi kullanılarak belirlenirken eritrosit yükseltgenmiş glutatyon (GSSG) ve indirgenmiş glutatyon (GSH) miktarları ise yüksek performanslı sıvı kromatografisi ( HPLC ) ile belirlendi.Tip 2 diyabet teşhisi almış kişilerin GSSG miktarları kontrol grubuna göre yüksek iken, GSH miktarlarının ise düşük olduğu gözlendi (p<0.005).Aynı şekilde diyabetiklerin hem eritrosit GSH-Px hemde GSH-Rd aktiviteleri kontrol grubuna göre düşük olduğu tespit edildi (p<0.005).Sonuç olarak tip 2 diyabetiklerin GSSG miktarı, GSH-Px ve GSH-Rd aktivitelerindeki artış ile GSH miktarında ve azalma oksidatif stres ile açıklanabilir.Anahtar Kelimeler: GSH, GSSG, GSH-Px, GSH-Rd, Tip 2 diyabet

In this study, erythrocyte glutathione peroxidase (GSH-Px) and glutathione reductase (GSH-Rd) enzymes activity of 60 people diagnosed with type 2 diabetes and 21 healthy persons as control group were determined using UV-spectrophotometer and erythrocyte oxidized glutathione (GSSG) and reduced glutathione (GSH) amounts were determined by high performance liquid chromatography (HPLC).While GSSG amounts of people diagnosed with Type 2 diabetes were higher than control group, amounts of GSH were found to be lower than the control group (p<0.005).In the same way, both GSH-Px and GSH-Rd activities in erythcrocytes of type 2 diabetes were found to be lower than the control group (p<0.005).In conclusion, it can be said that, increase of GSSG amount, GSH-Px and GSH-Rd activities with decrease of GSH amount of type 2 diabetics can be expressed by oxidative stress.Keywords: GSH, GSSG, GSH-Px, GSH-Rd, Type 2 diabetes
