HİSTOPATOLOJİK OLARAK UTERİN LEİOMYOM VE ENDOMETRİAL POLİP TANISI KONULAN OLGULARDA GHRELİN VE OBESTATİN'İN SERUM VE DOKU DÜZEYİNDE ARAŞTIRILMASI

Abstract

Myoma uteri; uterusun en sık rastlanan benign tümörüdür. Endometrial polipler, endometrial dokudan köken alan, içinde farklı sayıda bez, stroma ve kan damarı içeren, üzeri epitelle kaplı lokal büyümelerdir.Grelin, GHS-R ve grelin protein mRNA endometriumda ve myometriumda bulunduğunun gösterilmesi bu peptidlerin uterin leiomyom ve endometrial polip te eksprese edilip edilmediği myom komşuluğundaki myometrium ile polip komşuluğundaki endometrium dokusu arasında farklılık olup olmadığı, yine bu peptidlerin serum düzeylerinin polip ile myom grubu arasında farklı olup olmadığını uterin leiomyom ve endometrial polip fizyopatolojisinde olası rollerini araştırmak için bu çalışmayı planladıkÇalışma histopatolojik olarak uterin leiomyom tanısı konulan 45 olgu ile endometrial polip tanısı konulan 15 olgu üzerinde yürütüldü. Kontrol grubu olarak myom ve polip komşuluğundaki normal myometrium ile normal endometrium dokusu seçildi. Tüm gruplardaki dokular human anti grelin ve obestatin antikorları ile ayrı ayrı immünohistokimyasal yöntemle boyandı, Enzim immunoassay (EIA) ve ELİSA yöntemiyle çalışıldı.. Açil grelin, desaçil grelin, total grelin ve obestatin düzeylerinin gruplar arası karşılaştırılması yapılarak immunohistokimyasal boyama ile korelasyonu araştırıldı.Sonuç olarak; Bu çalışma ile grelin ve obestatinin hem endometrial polipte hemde uterin leiomyomda eksprese edildiğini ilk kez ortaya koyduk. Bu iki hormonun endometrial polip ve myom etyopatogenezinde olası rollerinin aydınlatılması için daha geniş klinik çalışmalara ihtiyaç olduğu bir gerçektir.Anahtar Kelimeler: Uterin leiomyom, endometrial polip, ghrelin, obestatin

Investıgatıon Of Plasma And Tıssue Ghrelın And Obestatın Levels In Hıstopatologıcally Dıagnosed Uterıne Leıomyoma And Endometrıal PolypMyoma uteri is a common benign tumour of the uterus. Endometrial polyps are local epithelial cooted growths that are originating from endometrial tissue and containing different amount of gland, stroma and blood vessels.We designed this study to determine the appearence of ghrelin, GHS-R, ghrelin protein m-RNA in endometrium & myometrium and the tissue around endometrium and myometrium. Also we examine the levels of these peptids in the serum of polyp and myometrium groups and wanted to investigate the role of these peptides in the pathophysiology of polyps and myomas.This study is due on 45 patients with leiomyom and 15 patients with endometrial polyp. Normal myometrium and endometrium are used for the control group Tissues in all groups are dyed by anti-ghrelin and anti-obestatin anticores. Also the tissue and serum of all cases are examined by enzyme immunoassay (EIA) and ELISA methods. The levels of acil ghrelin, desacil ghrelin, total ghrelin and obestatin are examined between groups and correlation between immunohystochemical dye is made.As a result we here in demostrated fort the first time that ghrelin and obestatin are expressed in both endometrial polyp and uterin leiomyoma. More clinical stıdies are needed to highlihght the role of these to hormones in the etiopathogenes of endometrial polyp & uterin myomas.Keywords; Uterin leiomyoma, endometrial polyp, ghrelin, obestatin
