DENEYSEL DİYABETİK FARE BEYİN DOKUSUNDAKİ APOPTOTİK DEĞİŞİKLİKLER ÜZERİNE D3 VİTAMİNİNİN ETKİSİNİN ARAŞTIRILMASI

Diabetes Mellitus (DM), insülin sekresyonu, insülinin etkisi veya her ikisindeki bozukluklardan kaynaklanan, hiperglisemi ile karakterize kronik, metabolik bir hastalıktır. Uzun süreli diyabette nörokimyasal, elektrofizyolojik, yapısal ve kognitif düzeyde serebral bozukluklar meydana gelmektedir. Vitamin D3, vitamin D`nin aktif metabolitidir ve değişik biyolojik olaylarda rol oynamaktadır. Son yıllarda vitamin D3'ün antioksidatif ve nöroprotektif özelliklere sahip olduğu gösterilmiştir.Bu çalışmada, streptozosin (STZ) ile oluşturulan deneysel diyabet modelinde vitamin D3`ün, fare beyin dokusundaki apoptotik değişiklikler üzerine koruyucu etkileri incelenmiştir.Çalışmada, 21 adet 30 ? 7 gr ağırlığında, 8 haftalık Balb/C türü erkek fareler kullanıldı. Tüm gruplar deney süresince standart fare yemi ve çeşme suyu ile beslendi. Deney hayvanları her grupta 7 hayvan olacak şekilde 3 gruba ayrıldı. Kontrol grubuna (Grup I) herhangi bir uygulama yapılmadı. Diğer 2 gruba 180 mg/kg olacak şekilde tek doz STZ 0, 1 M sodyum-sitrat tamponunda (pH: 4, 5) çözdürülerek i.p olarak uygulandı. Deneysel diyabet oluştuktan sonra diyabetik grup (Grup II) belirlenip herhangi bir uygulama yapılmadı. Diyabet + vitamin D grubuna (Grup III) 12 hafta süreyle vitamin D (50 IU/kg/gün) oral yolla verildi. Deney sonunda fareler dekapite edilip beyin dokuları çıkarıldı. Histolojik çalışma için rutin ışık mikroskobu takibi yapılarak dokular parafin bloklara gömüldü. Bloklardan alınan kesitlere histolojik, immünohistokimyasal ve TUNEL boyama yapıldı. Biyokimyasal çalışma için örnekler -80Co'de saklandıBiyokimyasal incelemelerde Grup II'de beyin dokularında total oksidan durum ve oksidatif stres indeksinde anlamlı bir artış vardı. Grup III'de bu değerler Grup I'e yakın gözlendi. Histolojik incelemelerde, Grup II'de astrositlerde yer yer proliferasyon yanısıra hücresel pleomorfizm ve hiperkromazi artışı vardı. Grup III'de Grup I'e benzer morfolojik özellikler mevcuttu. İmmünohistokimyasal boyamada; Grup II'de bax immünreaktivitesinde belirgin bir artış vardı. Grup III'de ise Grup I'e benzer immünreaktivite izlendi. TUNEL boyamada Grup II'de TUNEL pozitif hücrelerde anlamlı bir artış olmasına rağmen Grup III'de Grup I'e yakın boyanma görüldü.Sonuç olarak, DM un oluşturduğu hücre hasarına karşı vitamin D`nin koruyucu etkilerinin gösterilmesi, diyabetin komplikasyonlarını önlemek amacıyla vitamin D ile ilişkili tedavi yaklaşımlarının denenmesinin yararlı olabileceği ve bu konuda ileri araştırmaların yapılması gerektiği kanatine varılmıştır.Anahtar Kelimeler: Diabetes mellitus, vitamin D, beyin, apoptozis.
Diabetes mellitus (DM) is a hyperglycemic metabolic disorder caused by the lack of insulin secretion and/or effect of insulin or both of them. Electrophysiological, structural and cognitive cerebral disorders occur in the long-term diabetes. Vitamin D3, active metabolite of vitamin D, has important roles in the different biological events. In recent studies, it is determined that vitamin D3, has antioxidant and neuroprotective properties.In this study, it is investigated the protective effects of vitamin D on apoptotic changes in mice brain whith streptozotocin (STZ) - induced experimental diabetes.Twenty one Balb / C type male mice (aged 8 week and weighing 30 ± 7 gr) were used in this study. All of mice were received standard chow and water ad libitum during experimental period. Mice were randomly divided into three groups with seven animals per group. Mice in group I were used as control. Mice in group II and group III were injected with 180 mg/kg streptozocin solved in 0, 1 M phosphate-citrate buffer (pH:4, 5) intraperitoneally. After confirmed of experimental diabetes mellitus we separated the diabetes group (Group II) and no aplication was performed to this group for 12 weeks. In Group III mice were administered with 50 IU/kg/day vitamin D orally for 12 weeks. At the end of experimental study mice were decapitated and brain tissues were taken and some of them fixed in formaldehyde solution (10%). Tissues were processed to parafin wax by routine histological analysis, parafin sections were stained by immunohistochemically and TUNEL protocols. Remaining tissue samples were kept at -80Co until the biochemical analysis were performed.In Group II biochemical investigations showed us total oxidant status and oxidative stress index increased significantly. These parametres were much about the similar in Group I and Group III when compared each other. In histological examination, proliferation of astrocytes, cellular pleomorphism and hyperchromasia were observed in Group II. Brain structures of Group I and Group III were much similar. It was observed that expression of bax in brain tissues of Group II increased significantly. İmmunereactivities of bax were similar in Group I and Group III. Count of TUNEL(+) cells vere significantly higher in the Group II than cells of Group I and Group III.In conclusion, we suggest that vitamin D has protective effects againest the cell damage caused DM so, vitamin D therapy models can be improved by using different experimental research tecniques.Key Words: Diabetes mellitus, vitamin D, brain, apoptosis
