SODYUM METABİSÜLFİT İLE SODYUM NİTRİT'İN SACCHAROMYCES CEREVİSİAE ÜZERİNDEKİ METABOLİK ETKİLERİ

Gıda endüstrisinde yaygın olarak kullanılan katkı maddelerinin biyolojik sistemler üzerindeki etkilerinin değerlendirilmesi, insan sağlığı açısından büyük önem taşımaktadır. Bu çalışmada, gıda endüstrisinde yaygın olarak kullanılan koruyucu ajanlar arasında yer alan sodyum metabisülfit ve sodyum nitritin Saccharomyces cerevisiae üzerindeki metabolik etkileri incelenmiştir. Farklı konsantrasyonlarda (500 mg/L ve 1000 mg/L) uygulanan sodyum metabisülfit (SM) ve sodyum nitritin (SN), Saccharomyces cerevisiae'nin lipofilik vitaminler, antioksidan enzim düzeyleri, yağ asidi bileşimi ile malondialdehit (MDA), indirgenmiş glutatyon (GSH) ve oksitlenmiş glutatyon (GSSG) düzeyleri üzerindeki etkileri değerlendirilmiştir. Deneysel tasarım kapsamında kontrol, SM1, SM2, SN1 ve SN2 grupları oluşturulmuştur. Elde edilen bulgulara göre, toplam protein düzeylerinde kontrol grubuna kıyasla azalma olduğu belirlenmiştir. MDA düzeylerinde ise kontrol grubuna göre genel bir artış gözlenmiş olup, bu artışın özellikle SN1 ve SN2 gruplarında daha belirgin olduğu saptanmıştır. GSH miktarının kontrol grubuna kıyasla istatistiksel olarak anlamlı bir değişim göstermediği belirlenirken, GSSG miktarının SM1, SM2 ve SN2 gruplarında azaldığı tespit edilmiştir. Antioksidan savunma sistemine ait süperoksit dismutaz (SOD), katalaz (CAT), glutatyon peroksidaz (GSH-Px) ve glutatyon S-transferaz (GST) enzim aktivitelerinin, kontrol grubuna göre özellikle SN1 ve SN2 gruplarında arttığı belirlenmiştir. Yağ asidi analizi sonuçlarına göre, laurik asit miktarının SM gruplarında arttığı, SN gruplarında ise azaldığı saptanmıştır. Stearik asit miktarının kontrol grubuna kıyasla tüm deney gruplarında artış gösterdiği belirlenmiştir. Lipofilik moleküllerin gruplar arasındaki değişimleri incelendiğinde, SN1 ve SN2 gruplarında ?-tokoferol ve K1 vitamini düzeylerinin arttığı tespit edilmiştir. Sonuç olarak, bu çalışma sodyum metabisülfit ve sodyum nitrit uygulamalarının Saccharomyces cerevisiae hücrelerinde oksidatif stres oluşturduğunu ve lipid peroksidasyonunu artırdığını ortaya koymaktadır. Özellikle sodyum nitrit uygulanan gruplarda MDA düzeylerinin daha yüksek olması, bu maddenin oksidatif stres oluşturma potansiyelinin daha belirgin olduğunu düşündürmektedir. Antioksidan enzim aktivitelerinde gözlenen artışlar, hücrelerin oksidatif strese karşı savunma mekanizmalarını aktive ettiğini göstermiştir; ancak bu savunma yanıtlarının oluşan oksidatif hasarı tamamen engelleyemediği sonucuna varılmıştır. Bu çalışma, sodyum metabisülfit ve sodyum nitritin hücresel düzeyde oksidatif stres oluşturma potansiyelini ortaya koymakta ve gıda katkı maddelerinin biyolojik etkilerinin değerlendirilmesine katkı sağlamaktadır. The evaluation of the effects of food additives widely used in the food industry on biological systems is of great importance in terms of human health. In this study, the metabolic effects of sodium metabisulfite and sodium nitrite, which are commonly used as preservative agents in the food industry, on Saccharomyces cerevisiae were investigated. The effects of sodium metabisulfite (SM) and sodium nitrite (SN), applied at different concentrations (500 mg/L and 1000 mg/L), on lipophilic vitamins, antioxidant enzyme levels, fatty acid composition, and malondialdehyde (MDA), reduced glutathione (GSH), and oxidized glutathione (GSSG) levels in Saccharomyces cerevisiae were evaluated. The experimental design included control, SM1, SM2, SN1, and SN2 groups. According to the obtained results, total protein levels were found to decrease compared to the control group. A general increase in MDA levels was observed relative to the control group, with more pronounced elevations particularly in the SN1 and SN2 groups. While no statistically significant difference was detected in GSH levels compared to the control group, GSSG levels were found to decrease in the SM1, SM2, and SN2 groups. The activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSHPx), and glutathione S-transferase (GST), were determined to increase, especially in the SN1 and SN2 groups, compared to the control group. According to the fatty acid analysis results, lauric acid levels increased in the SM groups, whereas a decrease was observed in the SN groups. Stearic acid levels were found to increase in all experimental groups compared to the control group. Examination of changes in lipophilic molecules among the groups revealed that ?-tocopherol and vitamin K1 levels increased in the SN1 and SN2 groups. In conclusion, this study demonstrated that sodium metabisulfite and sodium nitrite applications induced oxidative stress and increased lipid peroxidation in Saccharomyces cerevisiae cells. The higher MDA levels observed particularly in the sodium nitrite–treated groups suggest a more pronounced oxidative stressinducing potential of this compound. Although the observed increases in antioxidant enzyme activities indicated the activation of cellular defense mechanisms against oxidative stress, these responses were insufficient to completely prevent oxidative damage. This study contributes to the evaluation of the biological effects of food additives by revealing the oxidative stress–inducing potential of sodium metabisulfite and sodium nitrite at the cellular level.
