SJÖGREN SENDROMUNDA SİGLEC DÜZEYLERİ

Abstract

Siglec'ler (Sialic acid-binding immunoglobulin-like lectins) esas olarak sialik aside bağlanarak etki gösteren hücre yüzey proteinleridir. İmmünglobulin benzeri lektin ailesine aittirler. Sıklıkla bağışıklık hücrelerinin yüzeyinde bulunurlar. Siglec'lerin özellikle doğal ve kazanılmış immünitenin düzenlenmesinde rol aldıkları bilinmektedir. Takipli 30 pirimer sjögren sendromu (PSS), 30 sistemik lupus eritematoz (SLE) tanılı hasta ve 30 sağlıklı kontrol gurubu çalışmaya dahil edildi. PSS, SLE ve kontrol gurubundan oluşan toplam 90 katılımcıdan siglec-1, siglec-2, siglec-10 düzeylerini ölçmek üzere serum ve tükürük örnekleri alındı. ELISA ölçümleri kitlerin kataloglarında belirtilen çalışma prosedürlerine uygun olarak çalışıldı. Veriler uygun istatiksel analiz kullanılarak değerlendirildi ve p<0.05 istatiksel olarak anlamlı kabul edildi. Ayrıca PSS tanılı ve çalışmaya alınan hastaların daha önceden yapılmış ve patoloji arşivinde bulunan tükürük bezi doku örneklerinden siglec-1, siglec-2, siglec-10 ekspresyonları immünohistokimyasal boyanarak değerlendirildi. Çalışmaya dahil ettiğimiz gruplar arasında yanlızca tükürük siglec-10 değeri açısından anlamlı farklılık olduğu görülmüştür (p=0.006). Bu farklılığın ise SLE grubu ile kontrol grubu arasındaki farktan kaynaklandığı ve SLE grubunun değerinin kontrol grubunun değerinden yüksek olduğu belirlenmiştir (SLE-kontrol için p=0.005). Pirimer sjögren sendromlu hastalarda Chisolm Masson'a göre yapılan patolojik grade ve serum siglec değerleri açısından anlamlı farklılık görülmemiştir (p >0.05). Pirimer sjögren sendromlu hastaların histopatolojik değerlendirme sonucu ile serum ve tükürük siglec değerleri ile aktivasyon indeksi değeri açısından anlamlı farklılık olmadığı bulunmuştur (p >0.05). Sonuç olarak siglec düzeylerinin immünite kökenli hastalıklarda değişebileceği ancak daha detaylı ve geniş kapsamlı çalışmalara ihtiyaç olduğu kanaatine varılmıştır. Siglecs (Sialic acid-binding immunoglobulin-like lectins) are cell surface proteins receptors by binding mainly to sialic acid. They belong to immunoglobulin-like lectin family. They are often found on the surface of immune cells. Siglecs are known to play a role in the regulation of innate and adaptive immunity. Thirty followed-up patients with primary Sjogren's syndrome (PSS), 30 patients with systemic lupus erythematosus (SLE) and 30 healthy control group were included in this study. In order to measure Siglec-1, Siglec-2 and Siglec-10 levels, serum and saliva samples were taken from a total of 90 participants consisting of PSS, SLE and control group. ELISA measurements were performed in accordance with the operating procedures specified in the kits' catalogs. Data have been evaluated using appropriate statistical analysis and p<0.05 was considered statistically significant. In addition, siglec-1, siglec-2 and siglec-10 expressions were evaluated by immune histochemical staining from salivary gland tissue samples from the patients who diagnosed with PSS and included in the study. It was observed that there was a significant difference between the groups we included in the study only in terms of the Siglec-10 value of saliva (p=0.006). It was determined that this difference was due to the difference between the SLE group and the control group, and the value of the SLE group was higher than the value of the control group (p=0.005 for SLE-Control). According to Chisolm Masson Grading, no significant difference was observed in patients with Primer sjögren's syndrome in terms of pathological grade and serum siglec values (p >0.05). It was found that there was no significant difference between the results of histopathological evaluation of patients with Primer Sjögren's syndrome and serum, saliva, siglec values and activation index value (p >0.05). As a result, it was concluded that siglec levels may change in immune-related diseases, but more detailed and comprehensive studies are needed.
