HİSTOPATOLOJİK OLARAK ENDOMETRİOZİS TANISI KONULAN OLGULARDA GRELİN VE OBESTATİN'İN SERUM VE DOKU DÜZEYLERİNİN ARAŞTIRILMASI

Abstract

Endometriozis, fonksiyonel endometrial bezlerin ve stromanın uterusdaki normal anatomik lokalizasyonundan farklı bir lokalizasyonda olması olarak tanımlanır.Ghrelin, GHS-R ve ghrelin protein mRNA endometriumda bulunduğunun gösterilmesi bu peptidlerin histopatolojik olarak endometriozis tanısı konulan olgularda ektopik endometriumda eksprese edilip edilmediği, serum düzeylerinde farklılık olup olmadığı endometriozis fizyopatolojisinde olası rollerini araştırmak için bu çalışma planlandı.Çalışma histopatolojik olarak endometriozis tanısı konulan 30 olgunun ektopik ve ötopik endometrium dokuları ile pelvik patolojisi olmayan 15 olgunun, ötopik endometrium dokularında ghrelin ve obestatinin immunohistokimyasal ve ELİSA yöntemi ile yine bu olguların serum düzeylerini araştırdık.Endometriozisli olguların ektopik ve ötopik endometriumlarında, kontrol grubu olguların ötopik endometriumlarında grelin ve obestatin her iki yöntem ile eksprese edildiği, grup içi karşılaştırmada sekretuvar fazda proliferatif faza göre ekspresyonun anlamlı bir şekilde daha fazla eksprese edildiği gösterilmiştir. Gruplar arası doku ve serum düzeylerinde anlamlı farklılık bulunmamıştır.Endometriozisli hastaların hem ötopik hem de ektopik endometrium dokularında ghrelin ve obestatin ekspresyonu gösterilmiştir. Bu bulgular endometriozisin etyolojisinin normal endometrium olabileceğini göstermiş olup daha ileri çalışmalarla endometrium reseptivitesinin belirlenmesine yeni bir yön verebilir.Anahtar kelimeler: Endometrium, endometriozis, ghrelin, obestatin

Researching Serum And Tissue Level Of Ghrelin And Obestatin İn The Cases Histopathologically İdentified As EndometriosisEndometriosis is defined as having different localization apart from normal anatomic location and functional endometrial in uterus.Now that it was showed that ghrelin is found at GHS-R and ghrelin protein mRNA endometrium, this study was planned to research whether these peptides are expressed in ectopic endometrium of phenomenon identified as endometriosis, serum level and their possible role in endometriosis physiopathology.In this study we, histopathologically, investigated the serum levels of these cases, the ectopic and eutopic endometrium tissues of 30 cases diagnosed as endometriosis and 15 cases without pelvic pathology, together with the ELISA and immunohistochemistry methods in the eutopic endometrium tissues of ghrelin and obestatin.In the ectopic and eutopic endometriums of the tissues with endometriosis, it was shown that ghrelin and obestatin was expressed together with both of the methods in the eutopic endometriums of the control group cases and compared to the proliferative phase, the expression in the secretory phase was much higher in the comparision of these internal groups. A meaningful variance has not been detected in the tissue and serum levels of these two groups.Both in the eutopic and ectopic endometrium tissues of the patients with endometriosis, ghrelin and obestatin expression was detected. Therefore, these findings have revealed that the etiology of endometriosis will probably be normal endometrium and might shape a new direction to the definition of endometrium reseptivness in the further advanced studies.Key words: Endometrium, endometriosis, ghrelin, obestatin
